inhibitor of the lipid peroxidation induced by iron salts and ascorbic acid.7) It has been revealed that mammalian cells in culture do not always Tequire transferrin, ifiTon is provided as a complex with appropriate chelators.8) To design a protein-free medium for hybridomas, we studied the effect of ferric citratc as an alternative to transferrin on thc proliferation of hybridomas. Mouse hybridoma IA-4-) producing IgG2u specific to akabane virus was used in this study. The hybridoma was a fusion product between the mouse myeloma cell linc, P3-X63-Ag8-Ul.iO) and BALBic spleen cells. Differential growth-inhibitory effects of gallium on B-lymphocyte lines in high versus low iron concentrations.
The growth inhibitory effects of gallium on a murine and human B-cell line were studied using two different serum-free culture systems: (a) ferric citrate medium containing 500 microM iron and (b) transferrin medium containing 5 micrograms/ml of iron-saturated transferrin (0.125 microM iron). For the human cell line in ferric citrate medium, 50% growth inhibition achieved in the presence of tra...
متن کاملThe Effect Of Vero Cells on Preimplantation Mouse Embryo in Cultur Medium
SUMMARY To study the effect of vero cells on preimplantation mouse embryos two groups of experiment were carried out. In experiment 1, pronuclear stage mouse embryos were co-cultured with Vero cells in RPMI medium and were examined under microscope for 4 days with 24h interval. The results showed co-cultured group developed better compared to control (87% to 80%, P<0.05) although, the rate ...
متن کاملPotential of Mouse and Human Embryonic Fibroblasts to Support Mouse Embryo Development Following Exposure to Laboratory Temperature
Purpose: Present study was designed to evaluate the potential of co-culture systems to overcome deleterious effect of exposure of mouse 2-cell embryos to low temperature. Materials and Methods: 2-cell embryos were flushed from oviduct of super ovulated NMRI mice into HTF medium with 15% BSA. After washing 3 times with HBSS and with 15% BSA, embryos were exposed to laboratory temperature (LT) (...
متن کاملP-36: The Effect of Lipoic Acid on Developmental Competence of Cultured Isolated Mouse Preantral Follicles
Background: α-Lipoic acid (LA) is a cofactor in mitochondrial multienzyme complexes and is well-known for its antioxidant properties in both of in vivo and in vitro condition. Materials and Methods: Preantral follicles were isolated from 2 weeks old NMRI mice ovaries. LA at concentrations of 0 and 100 μM was supplemented to culture media (α-MEM medium supplemented with 5% FBS, 100 mlU/ml FSH, 1...
متن کاملSerum-free Medium for Long-term Growth, Freezing, Cloning and Fusion of Myelomas and Hybridomas
Dear Editor: A number of laboratories and companies have reported on the development of serum-free media for the growth of myelomas and hybridomas in tissue culture (1-4,6-9). Due to the different growth requirements of different cell lines, these serum-free media generally have not been broadly applicable. For example, HB101 (Hana Biologies) supported the growth of SP2/0 cells, SP2/O-derived h...
